چکیده

Sheep erythrocytes have been spin labeled with 5-, 12-, and 16-nitroxystearic acid in order to investigate complement-induced changes in the physical state of the lipid bilayer. Formation of osmotic lesions in the membrane causes an increase in the fluidity of the membrane which overcomes the decrease in membrane fluidity caused by the interaction of the complement proteins. A decrease in membrane fluidity is observed only when complement-lysed membranes are resealed or when complement proteins react with isosmolar ghosts that do not undergo osmotic lysis. The decrease in bulk fluidity of the membrane is first observed when C8 binds to the membranes bearing C5b67 and is enhanced upon the subsequent binding of C9. The decrease in membrane fluidity shown by the electron spin resonance spectra of spin-labeled fatty acids suggests that certain of the complement proteins penetrate the membrane and interact with hydrophobic regions of the lipid bilayer.

Menadione-induced cytotoxicity effects on human erythrocyte membranes studied by electron paramagnetic resonance.

Menadione (2-methyl-1,4-naphthoquinone) is cytotoxic to hepatocytes. In order to begin to investigate the changes in the physical state of membranes induced by this cytotoxic substance, electron paramagnetic resonance (EPR) spin-labeling techniques were used in conjunction with spin labels specific for cytoskeletal proteins, bilayer lipids, or cell-surface sialic acid or galactose to investigat...

Electron spin resonance studies of erythrocytes from patients with myotonic muscular dystrophy.

Electron magnetic resonance experiments have demonstrated that spin-labeled myotonic erythrocyte membranes have spectra that are recognizably different from those of normal erythrocytes. The spin label incorporated in the erythrocyte membranes of patients having myotonic muscular dystrophy is apparently located in a less polar and somewhat more fluid region than the label in a normal membrane. ...

A spin label study of the effect of chrysotile asbestos on erythrocyte membranes.

Alterations in erythrocyte membranes caused by UICC B chrysotile asbestos fibres were studied in red cell ghosts using the spin label technique. The electron paramagnetic resonance (EPR) spectra of two sulphydryl reactive spin labels and one fatty acid spin probe in erythrocyte ghosts showed membrane protein modifications but no changes in lipid fluidity caused by the haemolytic chrysotile asbe...

MAGNETISATION AND ELECTRON SPIN RESONANCE STUDIES OF TETRAHEDRAL AMORPHOUS CARBON

The magnetisation and electron spin resonance (ESR) spectrum of two specimens of tetrahedral amorphous carbon (ta-C), deposited from a filtered cathodic arc, were measured over a wide temperature range. The magnetisation was found to consist of superparamagnetic, paramagnetic and diamagnetic contributions. The superparamagnetic contribution resembled that recently found in carbon prepared from ...

Effects of dehydroabietic acid on the physical state of cytoskeletal proteins and the lipid bilayer of erythrocyte membranes.

Dehydroabietic acid (DHAA) is a major aquatic toxic resin acid usually found in unbleached pulp mill effluents. This compound has been reported to accumulate in the red cells of rainbow trout and to cause hemolysis. To elucidate further understanding to the mechanism of action of this resin, the interaction of DHAA with human erythrocyte membranes has been monitored by electron paramagnetic res...
